我們設計2個研究聚焦在血糖變化和神經發炎以及藥物使用和睡眠結構改變來探討失智症的危險因子和相關致病機轉,第1個研究為探討血糖變化和神經發炎進一步導致失智症的可能機轉,我們利用急性外在葡萄糖濃度變化來檢驗對BV-2 微小膠細胞活性的影響去了解可能的致病途徑。我們選擇正常葡萄糖濃度(5.5mM)和高葡萄糖濃度(25mM)間的轉換來分析對微小膠細胞的細胞生長、氧化/發炎壓力、微小膠細胞活化、細胞凋亡和細胞自噬等變化,發現正常葡萄糖濃度轉變為高葡萄糖濃度為誘發微小膠細胞的氧化/發炎壓力、促進細胞生長、增加促發炎因子的分泌、壓力/發炎蛋白質的表現和增強LPS 誘發的發炎現象;而高葡萄糖濃度轉變為正常葡
