楊樹病蟲害調查及其防治

栽培在臺灣的楊樹(Populus spp.)目前罹患多種病害與蟲害,故進行相關調查及鑑定工作,對危害情況較嚴重之銹病和炭疽病,則以藥劑及精油進行防治及篩選。本研究期能藉由實務的探討,作為日後防治工作基礎,以解決臺灣楊樹在臺灣種植栽培上所面臨的病蟲害問題,來提昇其產量與品質。本研究首先調查雲林四湖、東勢、元長地區、臺大梅峰實驗農場四個地區,臺灣楊樹之病蟲害發生情況、種類,並採集病害發生部位及害蟲。再將採集發病部位之病葉及病根進行病原菌分離、純化及培養,除鑑定病原菌形態外,並進行分子鑑定、接種試驗,以完成柯霍氏法則(Koch’s postulate)。蟲害方面經調查、鑑定出危害之蟲害有:巨型天牛

、咖啡木蠹蛾(Zeuzera coffeae Niether)、小綠葉蟬(Jacobiasca formosana)、黃斑椿象(Erthesina fullo)、小白紋毒蛾(Orgyia postica)、大避債蛾(Clania preyeri Leech)、格斯特家白蟻(Coptotermes gestroi)、螺旋粉蝨(Aleurodicus disperses)及潛葉蛾Phyllocnistis citrella),而巨型天牛品種尚鑑定中。在病害鑑定方面,則根據形態特徵、真菌組織分離、分子鑑定,發現有銹病(Rust)、Alternaria tenuis、Ophiostoma quercu

s、炭疽病( Anthracnose)、藻斑病(Algal spot of poplar)、葉斑病(Leaf spot)、莖枯病(Stem blight)及灰黴病(Botrytis blight)。銹病菌Melampsora larici-populina (Kleb)及炭疽病病原菌Colletotrichum gloeosporioides (Penz.) Sacc之防治,選取市面上易購之殺菌劑二十九種及萃取自臺灣種植之木本植物之木材部和葉部精油共二十三種,進行防治並行篩選。試驗則測試銹病病原菌之孢子發芽率;炭疽病病原菌之病原菌菌絲生長抑制率和孢子發芽率。試驗結果顯示:對銹病病原菌孢子發芽抑

制效果較佳的藥劑,計有克收欣(Kresoxim-methyl)、白列克收欣(Boscalid + Kresoxim-methyl)、快得寧(Oxine-copper)、鉀碳酸氫鉀(Potassium hydrogen carbonate)、白列克敏(Pyraclostrobin + Boscalid);精油方面則有大葉桉(Eucalyptus robusta)葉子精油、澳洲茶樹(Melaleuca alternifolia)葉子精油、扁柏(Chamaecyparis obtuse)木材精油、紅檜(Chamaecyparis formosensis)木材精油、土肉桂(Cinnamomum osm

ophloeum)葉子精油、錫蘭肉桂(Cinnamomum zeylanicum)葉子精油、牛樟(Cinnamomum micranthum)葉子精油。有關對炭疽病病原菌孢子發芽抑制效果較佳的藥劑則有克收欣、殺紋寧(Hymexazol)、扶吉胺(Fluazinam)、撲克拉(Prochloraz)、鋅錳乃浦(Mancozeb);精油方面有大葉桉葉子精油、澳洲茶樹葉子精油、扁柏木材精油、紅檜木材精油、土肉桂葉子精油、錫蘭肉桂葉子精油,龍柏(Juniperus chinensis)葉子精油並再進行苗木防治試驗。所選取的五種藥劑和八種精油對銹病皆有預防性防治效果。其中克收欣、白列克收欣、檜木木材精油

中華蝠蛾之生物學特性及蛹蟲草接種家蠶之研究

本論文將分為兩個部份,第一部份為探討中華蝠蛾 (Endoclita sinensis) 之生物學特性。中華蝠蛾為鱗翅目、蝙蝠蛾科 (Lepidoptera:Hepialidae) 之昆蟲。本研究旨在觀察中華蝠蛾之生活史及生物學特性。研究結果顯示,該幼蟲會於林木之樹幹內蛀食孔道,且利用絲、碎屑及排泄物封閉蛀食孔口,並以蛀食孔周圍之植物癒傷組織為主要食物。在野外,其幼蟲呈聚落狀分佈,其終齡幼蟲雌蟲體長和體重,分別為86.5 ± 11.4 mm和6.5 ± 2.0 g,與雄蟲之體長和體重64.0 ± 12.8 mm、2.8 ± 1.3 g具有顯著性之差異。在毛序之分佈上,中華蝠蛾終齡幼蟲前

胸微毛區之D1、SD1和SD2形態特徵與Aenetus和Phassus屬同為蛀木性及以植物癒傷組織為食者相同。 中華蝠蛾之卵為圓形,表面具小刻點,直徑約0.8 mm,卵期30 ± 3天。在25℃、相對溼度100%時,孵化率達98.21 ± 1.32%。於25℃環境飼養下幼蟲共有6個齡期,各齡期之頭殼寬分別為0.3 ± 0.1、0.6 ± 0.1、1.7 ± 0.3、4.6 ± 0.6、6.8 ± 0.8、8.4 ± 0.3 mm。剛孵化之幼蟲並無啃食卵殼之行為,個體之間皆為單獨生活,會因競爭生長空間而有相互攻擊之現象。本研究發現中華蝠蛾幼蟲對含有血桐木粉成分之人工飼料,具有誘引取食之現象

。幼蟲化蛹前會於孔道中綴絲,絲為一層層與孔道垂直,且類似蜘蛛網狀之結構,幼將會於孔道底部化蛹,幼蟲期為294 ± 14天。中華蝠蛾幼蟲在21℃、24℃、27℃及30℃四種不同溫度與ADⅠ、ADⅡ、ADⅢ和ADⅣ四種不同人工飼料飼養下,其發育時間和體長均具有顯著性之差異,總幼蟲期為27℃中以ADⅢ人工飼料飼育之發育期為259.2 ± 3.8日最短,21℃中ADⅠ人工飼料飼育之發育期為321.0 ± 1.6日最長。中華蝠蛾蛹為圓筒形,長54.8 ± 5.2 mm。於蛹期時,可以由腹部末端之外性器分辨雌雄蟲。在羽化前蛹會利用旋轉蠕動之方式,將蛹體移至蛀食孔處羽化,蛹期為25 ± 4天。中華蝠蛾成蟲口

器退化,觸角絲狀且不發達,長0.5 ± 0.2mm。前翅基部具翅垂,其顏色為黃褐色,有許多深褐色之斑塊與黑色線條,中室有一塊白點;後翅為黑褐色,無其他翅紋。雄蟲體較細瘦,體長3.8 ± 0.5 cm,展翅5.8 ± 0.7 cm,後足之脛節上具有橘色之費洛蒙腺;雌蟲體較粗壯肥大,體長4.8 ± 0.8 cm,展翅8.9 ± 0.6 cm。成蟲平均壽命約為7天。本研究已可利用人工飼料餵飼中華蝠蛾幼蟲生長發育。中華蝠蛾成蟲之交配和產卵可能受光週期及環境溫度變化等因素影響,仍需再作進一步深入探討。 第二部份為蛹蟲草之生長特性及接種家蠶之研究,蛹蟲草 (Cordyceps militaris (

Vuill.)) 又稱北蟲草或北冬蟲夏草,為麥角菌科(Clavicipitaceae) 蟲草屬 (Cordyceps) 之昆蟲病原真菌 (entomopathogenic fungus)。本研究旨在探討人工誘導蛹蟲草子實體發生及生長所需之條件,並分析比較子實體與菌絲體之化學成分。研究結果顯示,採集自阿里山森林遊樂區塔山步道之蛹蟲草其菌絲之最適培養基為YMB培養基,最適培養溫度條件為23℃。利用蛹蟲草之菌絲培養液 (濃度82.5mg/ml) 做為接種原,以菌液注射方式接種,接種後進行蛹體覆蓋處理,移入23 ± 2℃光照10小時之環境下,並保持透氣維持80%之相對濕度,可成功誘導出子實體。

化學成分分析結果顯示,以蛹蟲草菌接種於家蠶蛹所形成子實體之多醣體含量163.51 ± 3.96 mg/g,及蟲草素含量98.20 ± 0.82 mg/g較菌絲體者為高;而腺苷含量則以冬蟲夏草菌絲體含量23.72 ± 0.08 mg/g較子實體者為高;胺基酸則以蛹蟲草菌接種於家蠶蛹所形成子實體之天冬氨酸含量31.820mg/g、麩氨酸含量41.263mg/g、及精氨酸含量17.31 mg/g 比人工培養基培育之蛹蟲草子實體者為高。
