平戶杜鵑開花物候研究—以臺大校園、金山、梅峰為例

植物物候如展葉或開花等生命週期性律動事件發生時間受環境相互影響的變化,是植物反應氣候變化最明顯的方式。平戶杜鵑為北臺灣公園綠地廣泛栽植之杜鵑品種,每年隨著杜鵑花季的到來,活動慶典爭相於盛花期舉辦,而近年隨著極端氣候及全球暖化的發生,是否會造成開花物候的改變值得進一步探討。目前國內物候研究面臨的挑戰之一為少有長期穩定且標準相同的物候觀測資料,取得相同標的在不同地區或長期累積之資料實屬不易。本研究目的為參考國際標準建立適用平戶杜鵑的物候生長階段量表及其觀測標準,做為物候觀察長期監測的依據並透過不同實驗地點的設計,配適物候溫時模式掌握平戶杜鵑開花物候配適情形及其配適條件,探討不同氣候條件對於物候溫

時模式影響。試驗中選定同品系同一批扦插生產之四年生盆植平戶杜鵑‘大紫’(Rhododendron ×pulchrum ‘Oomurasaki’ )植栽,於2020年9月頂芽轉變為花芽後,分別放置於三個試驗地:A臺灣大學校園(臺北市)、B金山杜鵑花苗圃(新北市)以及C臺灣大學梅峰農場(南投山區),進行不同氣候條件地區開花特性之觀察。平戶杜鵑花芽發育與前一年秋冬季及當年春季溫度有關,於2021年春季主要花期之始花日以金山苗圃最早(2月23日),其次是臺大校園(2月26日)而梅峰農場最晚(3月23日)。本研究中詳述了平戶杜鵑花芽發育外觀特徵並對照不同階段花芽內部花器發育情形。參考BBCH基本生長階段

為架構,建立「平戶杜鵑物候生長階段BBCH量表」,以提供平戶杜鵑物候階段更統一及標準化的比較方法。另以溫時模式增加設定不同的生長適溫閾值上限,進一步討論北臺灣兩試驗地花芽發育至開花所需要的生長度日積溫(Growing Degree Days, GDD)。結果以起始日期11月1日(自Stage 51可見花芽階段)累加日平均溫度(Tavg),設定基本溫度(Tbase)12.1℃及上限溫度(Tupper) 20℃之模式,計算2021年臺大校園及金山苗圃開花所需生長度日積溫(GDD)數值最接近且與花芽生長趨勢較相近,累加至始花日(Stage 61)約需352.22~365.56℃d,達盛花日(Stag

e 65)約需402.89~412.83℃d。以相同的模式方法回溯較早形成的花芽於臺大校園2020年12月提早開花日期以及金山苗圃前次花期2020年1月底始花日期之計算,也得到與實際開花相近日期區間,並得以推測2020年金山地區可能因1月上旬溫度較暖且日平均溫度多位在12.1~20℃有效區間而有較早開花之狀況。由於本研究中調查期間及樣本數有限,在開花模式選用及相關參數設定,尚須累積更長期物候觀測記錄,以確認模式的適用性再行調整修正,「平戶杜鵑物候生長階段BBCH量表」將有助於以標準化方式,提供未來物候觀察長期監測的依據。

‘滿天星’百香果莖頂組織培養之研究

本研究探討以不同鹽類濃度、培養基型態及生長調節劑,對‘滿天星’百香果莖頂初代及增殖培養之影響。鹽類濃度有1/4MS、1/2MS及MS,以1/2MS培養其培植體鮮重其雖較以MS培養小,但其癒傷組織率低,展葉良好。培養基型態有固態、雙相及液態紙橋培養基,其中液態紙橋培養基其培植體鮮重較重,且無癒傷組織,並展葉良好。生長調節劑有NAA(0~0.04 mg/L)、BA(0~0.4 mg/L)及NAA 0.02 mg/L與BA(0~0.4 mg/L)之組合,於各處理及組合以僅添加0.02 mg/L NAA,其培植體表現良好,鮮重可達15.4 mg,且無癒傷組織形成。初代培養之建立培植體取之季節,在

夏季摘取之培植體,不僅鮮重表現最佳,無癒傷組織生成。經本研究結果‘滿天星’百香果莖頂初代培養,建議培養基鹽類濃度以1/2MS、培養基型態以液態紙橋培養並添加 0.02 mg/L NAA其培植體且有較佳的生長情形。 增殖培養階段,在添加kinetin時,增殖率以0.6 mg/L kinetin結合0.02 mg/L NAA之固態培養基,其增殖率最佳達2.1個芽,其癒傷組織生成率極低。在添加2iP時,培植體伸長效果最佳,濃度0.4-1 mg/L 2iP結合0.02 mg/L NAA之固態培養基,芽長達5.4-5.8mm,但僅在低濃度之0.1 mg/L之2iP無癒傷組織生成,其他各濃度皆有癒傷組織

生成。增殖培養有添加TDZ之處理者皆有癒傷組織生成,且添加TDZ之處理者與其他兩種生長調節劑2iP及kinetin比較,其增殖及培植體伸長皆表現不佳。在溫度試驗增殖及培植體伸長三種溫度雖無差異,但在30。C不僅存活率最高,同時無褐化及癒傷組織生成。最後培養基型態以固態培養基較雙向培養基表現佳,雙向培養基之培植體顏色黯淡,且部分培植體出現玻璃質化的情況。
