近年來,隨著分子生物學和基因工程學的發展,抗體-藥物共軛物 (antibody-drug conjugate, ADC) 逐漸成為治療癌症的重要策略。市面上抗體-藥物共軛物大都為異質混合物,使其藥物代謝動力學、穩定性、耐受性等無法達到最佳效果;而臨床實驗所開發之均質抗體-藥物共軛物大多有藥物抗體比 (drug-to-antibody ratio, DAR) 低,進而使藥物效率差等問題。因此本研究中利用開發之亞硼酸-苯基磺醯基探針,將其專一性標記至人體免疫球蛋白 G (Immunoglobulin G, IgG) 之重鏈,以提供未來藥物的專一性位點修飾,再藉由修飾於探針的活性官能基團,以可逆加

成-斷裂鏈轉移 (reversible addition-fragmentation chain transfer, RAFT) 進行聚合反應,生成聚乙二醇 (PEGlyation) 高分子鏈及雙肽連接子 (dipeptide linker) 高分子鏈。隨後,以 10% 氫氧化鉀水溶液水解抗體-高分子鏈之酯基後,取其高分子鏈,藉由凝膠滲透層析儀 (gel permeation chromatography, GPC) 鑑定其長度。另設計以丹磺醯基連接硫醇 (Dansyl-SH) 之化學小分子,以模擬細胞毒性藥物美登素 (Mertansine, DM1),並將其共軛至雙肽連接子,以測試抗體聚合物

掛藥的可行性,期望藉由聚合反應使可掛藥位點增加進而提升藥物抗體比。最後藉由紫外-可見光光譜儀監測抗體-藥物共軛物之濃度變化,以測得藥物抗體比為 2.29。希望為後續生成均質的抗體-藥物共軛物提供良好的接枝策略,並帶來提高藥物抗體比的可行性。

淺海熱泉烏龜怪方蟹的行為模式與微生物特徵

台灣龜山島週邊海域有著世界罕見的淺海熱泉系統,泉口噴發著含有硫化氫的熱液。由於硫化氫會抑制生物的呼吸作用,對生物來說具有高度毒性。因此,高硫的極端條件,使龜山島的淺海熱泉系統呈現低度的生物多樣性。烏龜怪方蟹是淺海熱泉系統中的優勢種生物,其密度可達每平方公尺約有364隻。過去的研究指出:烏龜怪方蟹會受到含硫沉積物所吸引;此外,烏龜怪方蟹在淺海熱泉系統中的空間分佈數量,在不同月份間亦有差異。然而,目前並無研究文獻指出環境硫化物含量與烏龜怪方蟹空間偏好之關聯。因此,本研究運用行為實驗,探討來自不同季節的烏龜怪方蟹,對高硫環境的偏好程度。此外,亦針對烏龜怪方蟹的鰓組織及殼表進行菌相分析,以求瞭解菌相

組成對於烏龜怪方蟹生存於熱泉環境的可能關聯性。 在動物行為模式實驗部分,本研究在一長方形缸兩端分別加入已滅菌海水(C區)及含硫海水(S區),紀錄烏龜怪方蟹在裝置中5分鐘內的移動路徑,並針對在兩類海水間的初次和最終選擇、進入兩類海水區域的次數和累積逗留時間、以及相對位置等面向進行分析。實驗整體結果顯示:來自不同季節的烏龜怪方蟹,並無對高硫區域產生顯著的偏好性。此外,在動物組織的菌相分析方面,本研究針對二月與五月份烏龜怪方蟹的第五片鰓和殼表,進行DNA萃取與16s rRNA序列分析,結果顯示:鰓組織與殼表的微生物群落結構不同,但兩者同樣是以Campylobacteria為優勢種,次優勢種Ga

mmaproteobacteria在鰓上豐度相對偏高。推測烏龜怪方蟹能藉Campylobacteria與Gammaproteobacteria對硫化合物的氧化能力,將高毒性硫化合物氧化成低毒性硫化合物,這樣的菌相組成,可能有助於生存於高硫環境下的烏龜怪方蟹進行解毒機制。此外,烏龜怪方蟹殼表擁有豐富的Alphaproteobacteria,此類細菌能藉由不產氧的光合作用進行固碳,因此我們推測:殼表的菌群可能成為同伴的食物來源,進而吸引烏龜怪方蟹高密度地群居在淺海熱泉系統中。
