儀器分析在近百年內有顯著的巨大發展,質譜法 (MS) 也不例外。在過去數十年內,電子與電腦技術的興起改變了化學科學,化學科學不再是過去一個世紀的樣貌。過去十年內的大量化學相關研究因電子模組的投入而受益,像是微型控制器與微型電腦。在 第一章 中,呈現了過去數個世紀中,化學與電子科學協同演變的歷史過程,並介紹現代可實際用於化學研究的電子工具。接著是MS、蛋白質結構和MS與蛋白質研究關聯性三者之基礎原理說明。再進一步討論分析化學中,「自動化」所扮演的角色。此外,也對一利用化學鐘與震盪反應分析樣品之相對少見的分析化學策略進行評估。分析偵測系統能夠是質量靈敏、濃度靈敏或是同時兼具。舉例而言,紫外-可見

在心臟受到損傷所引發的修復重塑過程中,心肌纖維母細胞(cardiac fibroblasts)扮演重要的角色,包括myofibroblast型態改變、發炎前驅物細胞激素的分泌、膠原蛋白的合成、增加細胞外基質的代謝,進一步造成心臟的發炎。另外,在心臟發炎過程當中,發炎蛋白IL-6的產生,及心臟膠原母細胞轉型成myofibroblast表現α-smooth muscle actin 以增加自身的收縮表現。而這些反應都被認為是參與病理過程的調控,其含量也是心臟發炎的主要指標之一。Lysophosphatidylcholine (LysoPC) 已被證實累積於缺氧的心肌當中,且參與調控細胞的增生、平

的活性氧化物質透過,活化 JNK1/2路徑,導致轉錄因子NF-κB及FoxO1活化。另外,LysoPC所誘導的第二型環氧化酵素依賴性的IL-6表現,此實驗提供造成心臟纖維的治療目標。在第二部分,我們更進一步探討LysoPC所誘導下產生第二型環氧化酶所調控的路徑。在西方墨點與基因表現的結果發現,前處理抑制劑如PKCα (GÖ 6976)、粒線體ROS清除劑 (Mitotempo)、NADPH oxidase (DPI) 、JNK1/2 (SP600125) 、Drp1 (mdivi-1)或 FoxO1 (AS1842856) 可減少LysoPC誘導的COX-2蛋白和基因表現。而上述的抑制劑也可抑

制LysoPC所誘導下膠原蛋白的分泌。且在前處理選擇性第二型環氧化酶抑制劑(celecoxib及NS398)抑制膠原蛋白的表現。在前處理Mitotempo、GÖ 6976或SP600125 可抑制JNK1/2的磷酸化;此外,前處理Mitotempo或GÖ 6976可抑制PKCα的磷酸化前處理GÖ 6976可抑制Drp1絲胺酸616上的磷酸化。進一步,我們證明LysoPC誘導粒線體氧化壓力增加,且造成粒線體分裂及去極化。這一部分的,我們證明LysoPC誘導的第二型環氧化酶導致膠原蛋白的分泌,透過粒線體的活性氧化物質,活化PKCα/ Drp1/JNK1/2路徑,調控FoxO1絲胺酸256上磷酸化。

另外,在S256D FoxO1大量表現情況下,也可增加膠原蛋白的分泌,且增加的膠原蛋白可被第二型環氧化酶抑制劑所抑制。進一步探討,干擾RNA抑制EP4後抑制LysoPC誘導的膠原蛋白產生。這些結果證明,LysoPC誘導的第二型環氧化酶與第二型前列腺素的產生,進一步造成膠原蛋白產生,是藉由EP4受體調控。此實驗結果提供LysoPC誘導心臟纖化的機制,以提供減輕心臟纖維化的反應。
