目前與藻類上真菌多樣性相關之研究還在初期階段,只有少數研究其分佈及多樣性,特別是在海洋大型藻類上。細翼枝菜 (Pterocladiella capillacea)為北臺灣終年生長的優勢紅藻物種,本研究旨在調查生長於細翼枝菜上可培養之內/外生真菌多樣性,主要目標為:(1) 調查自基隆潮境公園採集之健康及死亡細翼枝菜上之真菌多樣性;(2) 比較分離菌株使用之培養基種類以及表面殺菌方式對細翼枝菜上真菌多樣性之影響;(3) 探討細翼枝菜上真菌之季節性變化。本研究自2013年5月至2014年5月期間,每兩個月至臺灣基隆潮境公園採集健康及死亡之細翼枝菜藻體。健康及死亡之細翼枝菜藻體片段以三種不同方式做表
面殺菌或漂洗:(1) 4%次氯酸鈉溶液;(2) 70%酒精;(3) 滅菌人工海水,並將其置於葡萄糖-酵母萃取物-蛋白腖瓊脂海水培養基 (GYPS)、馬鈴薯葡萄糖瓊脂海水培養基 (PDAS)及人工海水瓊脂培養基 (SA)上培養。將藻體片段上長出之真菌分離成為單一菌株,並依其菌落形態初步分群後,將其核糖體核酸 (rRNA)基因之內轉錄間隔區 (internal transcribed spacer, ITS)序列以美國國家生物技術資訊中心 (NCBI)資料庫中的BLAST比對鑑種。結果顯示分離頻率 (%),分離出真菌之藻體片段數量/總藻體片段數量×100)最高者為以滅菌人工海水漂洗處理,並培養於G
YPS的健康藻體 (93.86%);最低者為以4%次氯酸鈉溶液滅菌處理,並培養於SA的死亡藻體 (33.32%)。在健康藻體上分離出119種真菌,死亡藻體上分離出85種真菌。整體上共分離出3,187株129種真菌,其中子囊菌有119種,擔子菌9種,毛黴菌1種。細翼枝菜上出現率最高之優勢菌群為散囊菌目 (28.33%),其次為肉座菌目 (14.40%)、鎖擲酵母目 (13.62%)、煤炱目 (9.57%)、格孢菌目 (5.90%)及炭角菌目 (5.62%)。豐富度 (abundance)最高之菌種為擔子酵母菌Rhodosporidium fluviale,共分離出346株,佔10.86%;其次為
分離出213株之Aspergillus sp.-1,佔6.68%。不同季節中的結果顯示細翼枝菜上之真菌在溫度較高月份,其物種豐度 (species richness)及豐富度皆高於溫度較低月份。在不同分離培養基上 (SA、GYPS、PDAS)除整體出現率外,其物種豐度並未顯示有顯著的差異。未來應對細翼枝菜上無法培養之真菌其多樣性、生態定位及在宿主之定殖情形做進一步的探討。
