植物為了克服多種環境逆境,會引發各種的訊息調控,以增加抗性或適應性。先前的研究於水稻懸浮培養液中鑑定出分子量為34 kDa之蛋白質 (PR-8),且屬第三型幾丁質酶,並以各種逆境反應相關激素處理後發現,PR-8基因能受到甲基茉莉酸(MeJA)的誘導而表現。本研究進一步驗證PR-8基因在水稻幼苗全株皆會受MeJA之調控而有增加表現量。為了確認PR-8啟動子與JA訊息傳遞之調控活性區段,先以基因資料庫分析PR-8啟動子序列,發現有數個MYB、MYC、ERF和WRRY等逆境調控相關轉錄因子之結合域,依其分布初步將PR-8啟動子序列以5’端序列遞減方式設計四片段PR-8p.D0、D1、D2及D3構築

色藍色產物大量呈現在維管束,故推測GFP-GUS融合蛋白外泌至質外體後,累積於維管束周邊。綜合上述結果,PR-8啟動子D2-TATA間有1個MYB、2個ERF、2個MYC及2個WRKY反應之cis-elements,這些轉錄因子結合域可能與JA訊息傳遞有相關,且其基因中的5’UTR可能也影響啟動子的活性。未來可利用Osss分泌性及其啟動子誘導的特性,建立一具有應用性的水稻懸浮細胞分泌性外源蛋白質的表現系統。
