第二型糖尿病(Type 2 diabetes, T2D)是一種內分泌異常所導致的疾病,近年來已成為全球第九大死亡原因。T2D的患者常伴隨著許多併發症例如:慢性腎臟疾病 (chronic kidney diseases, CKDs)、心血管疾病 (cardiovascular diseases, CVDs)、高血壓及神經退化性疾病,其中造成T2D患者死亡的原因主要為CKDs及CVDs。Metformin目前是治療T2D的一線臨床用藥,許多研究證明了metformin可以改善CVDs及CKDs;研究顯示血小板 (platelet) 在CVDs和腦血管疾病 (cerebrovascular dis

eases) 中扮演著關鍵角色,對於止血和動脈血栓的形成亦至關重要的作用。根據研究指出metformin (1 mM) 可以透過活化protein kinase A顯著降低棕櫚酸 (palmitic acid) 刺激的人臍靜脈內皮細胞 (human umbilical vein endothelial cell, HUVEC) 功能障礙和抑制藉由ADP (adenosine diphosphate) 刺激大鼠的血小板活化及粒線體功能障礙 (mitochondrial dysfunction),顯見metformin對於生物體內的抗血小板效果卓越;因此metformin可能是治療動脈血栓形成的潛
